A MODEL OF SUBARACHNOID HEMORRHAGE IN RATS

AIM To build a simple and repeatable animal model of SAH. METHODS SAH was introduced by passing a nylon thread up through the right internal carotid artery and piercing a hone in the right anterior cerebral artery. At 12 and 24h, the rats were evaluated with rotarod test and the behavior scale (5-point scale). RESULTS The rats were trained through rotarod test and then randomly divided into three groups, including vehicle group treated with vehicle after SAH, nimodopine treated group (i.p 0.25mg/kg 5min, 6h, 12h after SAH ) and sham group.At the point of the perforation there was usually a capping clot. There was always blood in the basal cisterns with some spread over the hemisphere. The time of rotarod test at 12h was as follows (mean 卤 S.E.V): vehicle(n=5), 40卤22s; nimodopine(n=5), 121卤49s; sham (n=8), 260卤89s(P=0.001 overall; P=0.002, vehicle versus sham; P=0.043, nimodopine versus sham). The time of rotarod test at 24h was as follows(mean 卤 S.E.V): vehicle (n=5), 27卤18s; nimodopine(n=5), 73卤34s; sham(n=8), 184卤45s(P=0.03 overall; P=0.025, vehicle versus sham). The values for the 5-point scale at 12h were as follows(mean 卤 S.E.V): vehicle(n=5), 2.4卤0.5s; nimodopine(n=5), 1.6卤0.5s; sham (n=8), 0卤0s(P=0.000 overall; P=0.023, vehicle versus sham; P=0.007, nimodopine versus sham). The values for the 5-point scale at 24h were as follows(mean 卤 S.E.V): vehicle(n=5), 3.2卤0.8s; nimodopine(n=5), 1.8卤0.4s; sham(n=8), 0卤0s(P=0.000 overall; P=0.003, vehicle versus group; P=0.044, vehicle versus nimodopine). CONCLUSION We present a simple and reliable model of SAH in the rats which allows to evaluate novel compounds and new drugs for treatment of SAH.
